RESUMO

We described a challenge named "Diabetic Retinopathy (DR)-Grading and Image Quality Estimation Challenge" in conjunction with ISBI 2020 to hold three sub-challenges and develop deep learning models for DR image assessment and grading. The scientific community responded positively to the challenge, with 34 submissions from 574 registrations. In the challenge, we provided the DeepDRiD dataset containing 2,000 regular DR images (500 patients) and 256 ultra-widefield images (128 patients), both having DR quality and grading annotations. We discussed details of the top 3 algorithms in each sub-challenges. The weighted kappa for DR grading ranged from 0.93 to 0.82, and the accuracy for image quality evaluation ranged from 0.70 to 0.65. The results showed that image quality assessment can be used as a further target for exploration. We also have released the DeepDRiD dataset on GitHub to help develop automatic systems and improve human judgment in DR screening and diagnosis.

BACKGROUND: Nevus is very common; however, melanoma is slightly related to the deterioration of nevus because of its vulnerability to solarization, friction, aging, heredity, and other factors. Early diagnosis is essential for melanoma treatment, since patients have a high survival rate with early detection and treatment. Computer-aided diagnosis has been applied in the differential diagnosis of melanoma and benign nevi and achieved high accuracy, but it does not suit the screening of nevi because most studies are based on dermoscopy with a narrow field of vision and performed by professional doctors. Therefore, this study aimed to present the accuracy and effectiveness of our algorithm. METHODS: Based on whole-face images of patients, the authors used logistic regression and the Newton method to detect the nevus region. Then, Python and OpenCV were employed to detect the lesion edge and compute the area of the regions. A multicenter clinical trial with a sample size of 600 was then conducted to evaluate the effectiveness of the algorithm. RESULTS: The algorithm detected 2672 nevi from 600 patients, in which there were 195 patients of missed diagnosis and 310 patients of misdiagnosis. The Kappa value between 2 groups was 0.860 (>0.8). Paired t-test showed no significant difference between 2 groups' area results (P = 0.265, P > 0.05). CONCLUSION: Within the limitations of this study, the authors demonstrated a high agreement between algorithm's detection and doctor's diagnosis. Our new algorithm has great effectiveness in nevus detection, edge segmentation, and area measurement.

Mulberry-silkworm ecosystem is one of the important agro-ecosystems in China. Based on the principles and methods of emergy analysis, this paper studied the interior structure of mulberry-silkworm ecosystem and its relationship with exterior environment and economy. Some emergy indices for this ecosystem were quantitatively calculated, and compared with those of the agro-ecosystem in China. The results showed that the emergy investment ratio, emergy yield ratio, environmental loading ratio and emergy sustainability index was 3.78, 4.68, 0.18 and 26.0, respectively, suggesting the low environmental pressure and good ecological benefit in mulberry-silkworm ecosystem in China. Hi-technology was required to further decrease the labor force input and enhance the comprehensive utilization of sericultural resources.

Glioblastoma, the most aggressive primary brain tumor in humans, exhibits a large degree of molecular heterogeneity. Understanding the molecular pathology of a tumor and its linkage to behavior is an important foundation for developing and evaluating approaches to clinical management. Here we integrate array-comparative genomic hybridization and array-based gene expression profiles to identify relationships between DNA copy number aberrations, gene expression alterations, and survival in 34 patients with glioblastoma. Unsupervised clustering on either profile resulted in similar groups of patients, and groups defined by either method were associated with survival. The high concordance between these separate molecular classifications suggested a strong association between alterations on the DNA and RNA levels. We therefore investigated relationships between DNA copy number and gene expression changes. Loss of chromosome 10, a predominant genetic change, was associated not only with changes in the expression of genes located on chromosome 10 but also with genome-wide differences in gene expression. We found that CHI3L1/YKL-40 was significantly associated with both chromosome 10 copy number loss and poorer survival. Immortalized human astrocytes stably transfected with CHI3L1/YKL-40 exhibited changes in gene expression similar to patterns observed in human tumors and conferred radioresistance and increased invasion in vitro. Taken together, the results indicate that integrating DNA and mRNA-based tumor profiles offers the potential for a clinically relevant classification more robust than either method alone and provides a basis for identifying genes important in glioma pathogenesis.

Homeobox genes encode transcription factors notable for their ability to regulate embryogenesis. Here, we report the discovery of a cluster of 12 related homeobox genes on the X chromosome expressed in male and female reproductive tissues in adult mice. These reproductive homeobox on the X chromosome (Rhox) genes are expressed in a cell type-specific manner; several are hormonally regulated, and their expression pattern during postnatal testis development corresponds to their chromosomal position. Most of the Rhox genes are expressed in Sertoli cells, the nurse cells in direct contact with developing male germ cells, suggesting that they regulate the expression of somatic-cell gene products critical for germ cell development. In support of this, targeted disruption of Rhox5 increased male germ cell apoptosis and reduced sperm production, sperm motility, and fertility. Identification of this family of homeobox genes provides an opportunity to study colinear gene regulation and the transcriptional control of reproduction.
